Synthesis in Haemolysed Erythrocytes

چکیده

1. A preparation of completely haemolysed erythrocytes from the blood ofdomestic fowls, which actively incorporates [Ox-14C]glycine into haem, is described. 2. The effects of time and of glycine concentration on the rate of synthesis were studied. 3. Stimulation of the synthesis up to sixfold was obtained with some preparations of boiled yeast extract. 2:4-Dinitrophenol inhibited the synthesis almost completely at 5 x 10-4M. 4. Synthesis of free protoporphyrin also occurs during incubation of the haemolysate. 5. Contrary to the findings of Gajdos & GajdosTorok (1951), using the blood of phenylhydrazineanaemic rabbits, sodium fluoride was found to inhibit free porphyrin synthesis both in haemolysates and in whole blood of fowls. 6. The effects of various other substances, including some erythropoietic agents, are also described.

A 30 week gestation infant developed necrotising enterocolitis associated with Clostridium perfringens septicaemia at 3 weeks of age. He responded to treatment with intravenous fluids, antibiotics, and blood, but his blood haemolysed. Because of anaemia further blood was given, but, within minutes he died. Examination of his red cells showed an increase in T activation.
